This antibody recognizes a sialoglycoprotein of 39 kDa, identified as glycophorin A (GPA). It is present on red blood cells (RBC) and erythroid precursor cells. It has been shown that glycophorin acts as the receptor for Sandei virus and parvovirus. Glycophorins A (GPA) and B (GPB), which are single, trans-membrane sialoglycoproteins. GPA is the carrier of blood group M and N specificities, while GPB accounts for S and U specificities. TMB is a noncarcinogenic substitute (Ames test negative) for benzidine suitable as peroxidase substrate for use in ELISA procedures. The substrate produces a soluble end product that is pale blue in color and can be read spectrophotometrically at 370 or 620-650nm. The TMB reaction may be stopped with 2M H2SO4 (resulting in a yellow color), and read at 450nm. A sensitive and specific reagent for the detection of blood, assay of haemoglobin and assay of horseradish peroxidase (HRP).

TMB Membrane Substrate develops an insoluble, permanent dark blue reaction in the presence of peroxidase. The substrate is intended for western blot, dot blots, and line assays and offers comparable performance and sensitivity to some chemiluminescent substrates. This substrate is not recommended for microwell or immunohistochemical applications. Stable for 2 years, from the date of manufacture, when stored at room temperature (15 to 25C) or 48 months when stored at 2-8C.
